چکیده

Background Robust and reliable dynamic bioreactors for long term maintenance of various tissues at milliliter-scale on the basis of a biological, vascularized matrix (BioVaSc) have been developed at the Fraunhofer IGB in Stuttgart, Germany. As an intestinal in vitro equivalent, seeding of the matrix with CaCo-2 cells yielded in the self-assembly of a microenvironment with the typical histological appearance of villus-like structure and morphology [1]. We modified this matrix (BioVaSc) cell (CaCo-2) system to some extent with the aim to develop 3D intestinal equivalents for systemic preclinical testing of orally applied drug candidates in microliter-scale on a human Multi-Organ-Chip (MOC), which consists of different organ equivalents important for ADMET (adsorption, distribution, metabolism, excretion, toxicity) testing.

Automated substance testing for lab-on-chip devices

Background A smartphone-sized multi-organ-chip has been developed by TissUse. This platform consists of a microcirculation system which contains several fully endothelial-cellcoated microchannels in which organ equivalents are embedded. Briefly, Human 3D organ equivalents such as liver and skin could be maintained functional over 28 days and treated with chemical entities in this microcirculati...

Integrating skin and vasculature in a Multi-Organ-Chip Platform

Background Tests for drug development require an almost perfect fit with the human (patho-) physiological microenvironment. The majority of skin equivalents currently commercially available are based on static culture systems emulating only human epidermis, or combining epidermis and dermis in so-called full thickness skin equivalents. None of the existing systems contain important elements, su...

Dynamic culture of human liver equivalents inside a micro-bioreactor for long-term substance testing

Background Current in vitro and animal tests for drug development are failing to emulate the organ complexity of the human body and, therefore, to accurately predict drug toxicity. In this study, we present a self-contained, bioreactor based human in vitro tissue culture test system aiming to support predictive substance testing at relevant throughput. We designed a microcirculation system inte...

Integrating biological vasculature into a multi-organ-chip microsystem.

A chip-based system mimicking the transport function of the human cardiovascular system has been established at minute but standardized microsystem scale. A peristaltic on-chip micropump generates pulsatile shear stress in a widely adjustable physiological range within a microchannel circuit entirely covered on all fluid contact surfaces with human dermal microvascular endothelial cells. This m...

Human multi-organ chips – a possible solution for animal free systemic ADMET testing

Animal free systemic toxicity and ADMe testing predictive of human exposure is currently the major challenge for science, regulatory bodies and the industry involved. On the one hand, modern test system engineering focuses mainly on single organ equivalents, rather than the systemic combination of organs. On the other hand, none of the currently available systems ensures long-term homeostasis o...
